Profile Summary

We use mathematical modeling and machine learning based strategies for patient selection in clinical trials. We utilize longitudinal data from interventional and observational studies, that the clients conduct, to reveal different endotypes. Clients include Bial Biotech and Roche I2O. Bial Biotech: - --Spearheaded ideation and development of a machine learning algorithm for feature engineering and adaptive clustering of dynamical time-series metabolomics data characterizing Parkinson’s patients’ response to a novel drug in Phase 1b clinical trials. Graph-theoretic approaches were used to identify clues for a fluxes in the metabolic network for lipids. Analyzed longitudinal metabolomic and symptomatic data from Michael J Fox's PPMI (Parkinsons progression markers initiative). --Identified groups of linear metabolite progressors that associate with specific symptomatic scores for Parkinsons disease. --Built a first of its kind stochastic model that indicated the extent of instabilities in Likert scale symptomatic scores for Parkinson's disease. Roche I20: --Developed a customized algorithm for clustering of dynamical symptomatic scores characterizing the prognosis of Ulcerative colitis post Etrolizumab administration. --Applied a customized random-forest based algorithm that identified with good recall, ulcerative colitis subjects that worsen post induction phase. The work can potentially help in deciding the continuation of Etrolizumab treatment and hence mitigate the treatment costs. --Implemented generative AI (Topic modeling) methodology and stability techniques therein to characterize associated disease trends in etrolizumab Phase III trials. --Explored the use of scRNA-seq analysis suites (Seurat, scanpy) coupled with trajectory reconstruction techniques.
