Profile Summary

In 2017 I returned to Seattle from San Francisco after working for Amgen for 19 years. I joined my Executive Director to create a biotechnology company in Seattle. I was the principal scientist in charge of biochemistry. I left in 2019 to form my own biotechnology company in Tacoma WA. I put together a small team in a science incubator across from the University of Washington - Tacoma campus. We provide protein science services to small and large pharmaceutical companies across the nation. Our growth has been small but steady in the time of the Covid epidemic. I joined the protein chemistry group at Amgen in Seattle in 2002. My area provided purification and formulation of biologics and protein reagents from various cell lines to support of Amgen research groups. I handled molecules surrounding the therapeutic targets including the antigens for SAR. Specializing in the purification development for mid-scale rodent and cyno surrogate IgG2 purifications. This included many forms of chromatography and various solutions. I am familiar with the use of Waters, AKTA Explorer, Express and Pilot systems, and protein characterization using Waters HPLC, Forte Bio, and Applied-Bio NTS. I have produced proteins from mammalian, microbial, yeast, baculo virus, and hybridoma expression. I joined the Process Development group at AMGEN in 1998 where I trained in the separation development of non-tagged biologics by conventional media techniques. I focused on the pre biologic therapeutic for pre PK / toxicology studies and the surrogate protein counterpart. Modalities included soluble receptors/ligands, Fc fusion protein, enzyme and antibody proteins. Scales of 1 – 100L fermentation in various host cell lines. I provided formulation strategies to meet goals of clean products with the lowest possible level of aggregate and host contaminants. These process strategies were then transferred to engineering colleagues to take the molecule to clinic. I started my professional career in the renal pharmacology clinical lab at Fred Hutchinson Cancer Research Center in Seattle. My time was divided between clinical assays of patient cyclosporin levels using MEIA and FIA and basic research in kidney disease. I spent my last year as laboratory manager with 1 assistant technician. I developed skills including HPLC, radiography, and biochemical assays. One of my strengths is an ability to see beyond the individual tasks and tie together key factors that assured the overall picture of the research problem. In college, I worked under Dr. D. Lightfoot in the Biotechnology Program at Eastern Washington University. I learned bacterial fermentation, including the transfection and growth parameters for bioprocessing. I also enjoyed the opportunity to create a hybridoma cell line to express anti beta galactose dehydrogenase antibodies using mouse splenocytes and NS0 cells. I provided monoclonal antibodies for an internal biological study. In the process I became familiar with immunochemistry and animal handling techniques that would serve me well at the Fred Hutch and throughout my career.
