Profile Summary

Technical Competencies: 1) Chromatography – I am competent in the use of Agilent, Shimadzu, and Empower HPLC systems. 2) Protein purification – I have used size exclusion, ion exchange and reverse phase chromatography to purify therapeutic candidate proteins, as well as antigens used in antibody production (Monash University Protein Production Unit). This involves the optimization of high throughput expression and purification processes while maintaining product quality. 3) Enzymology & Assay development – I have developed both fluorescence and HPLC based assays for metalloproteases. I am confident with the conduct of enzyme kinetic studies. 4) Tissue culture – Using mammalian and bacterial expression systems, I have generated antigens for antibody production, according to SOPs and GMP guidelines. 5) In vitro organ bath studies conducted using isolated tissues. 6) In vivo studies - Blood pressure measurements in anesthetised rats. Research Interests : 1) Isolation and characterisation of proteins in animal venoms that mediate various toxic effects following envenoming. 2) Determining the ability of currently available antivenoms to neutralise the effects of these toxins. 3) Enzymes that play a key role in cardiovascular and neurodegenerative disease with a particular emphasis on Endothelin converting enzyme and Neprilysin. 4) Components of the endothelin system as biomarkers of cardiovascular disease. 5) Identification of novel drug leads from animal venoms - with a particular emphasis on the discovery of molecules that effect ion channels and components of the endothelin pathway. Ongoing projects: 1) Deposition in the brain of a protein known as amyloid beta is a hall mark of Alzheimer's disease. Several enzymes degrade amyloid beta. I have discovered a peptide which stimulates two amyloid beta degrading enzymes. I am currently conducting a preclinical study on this peptide using a mouse model of Alzheimer's disease. 2) I have identified a protein in a venom which stimulates a specific type of Calcium channel. I am currently conducting the biochemical and pharmacological characterisation of this protein, with the aim of developing it as a valuable tool in biomedical research. 3) Determining how components of the endothelin system can serve as biomarkers of various disease states such as cancer and cardiovascular disease.
