I did my doctorate from CSIR-Imtech, Chandigarh, India and pursuing postdoctoral training at Purdue University, West Lafayette, United States. For my PhD thesis project, I worked on PknA, an essential eukaryotic-type serine/threonine kinase from Mycobacterium tuberculosis. I investigated role of juxta membrane domain towards its catalytic activity and phosphorylation mediated  activation mechanism of PknA. This required me to clone, express and purify the protein and its variants from Escherichia coli. The purified catalytic module of PknA was used for structural determination using X-ray crystallography  which gave us mechanistic understanding of PknA activation. Biochemical and biophysical techniques including kinase assays, western blotting and circular dichroism were utilized to further characterize the proteins. The findings from this study were published in FEBS journal. 
My research interests involve using structural biology and biochemical techniques to gain understanding as how dysregulated proteins cause disease. In postdoctoral period, I investigated regulation of P-Rex1, a guanine nucleotide exchange factor over-expressed in different cancers. I determined the structure of its regulatory domain, DEP1 using X-ray crystallography and elucidated its role in autoinhibition using fluorescence based enzymatic assays, Small Angle X-ray scattering and liposome sedimentation assays.
