Profile Summary

I gained a broad background in cell biology and biochemistry from my graduate work that I have advanced as a post-doctoral fellow. As a graduate student, I investigated the crosstalk between the Wnt signaling pathway and mRNA translational regulation under stress. My graduate studies focused on Stress Granule (SG) assembly under pharmacological stress conditions and did not give insight into which mRNAs were regulated or where this might occur within the cell. For my post-doctoral work, I joined the laboratory of Dr. Jeffery L. Twiss at University of South Carolina where I have advanced my expertise in SGs to work with neurons. The highly polarized neurons provided me an elegant system to study the SGs under normal physiological conditions. My studies focused on how mRNAs are stored in axons and how they come out of the storage depots in response to injury. My data showed that the SG protein G3BP1 is present in PNS axons and associates with some but not all axonal mRNAs. The protein is dynamic, showing assembly vs. disassembly into SGs depending on the growth state of the axons. Importantly, these changes in axon growth correlate with specific changes in intra-axonal protein synthesis. I also uncovered an endogenous pathway that sensory neurons use for disassembling axonal SGs. Specifically, Casein Kinase 2 (CK2)-dependent phosphorylation of axonal G3BP1 triggers disassembly of axonal SGs, releasing axonal mRNAs for translation and increasing axon growth. Temporal and spatial specificity of CK2a in axons is driven by translational regulation of its mRNA (Csnk2a1) in axons after injury. In summary, I have a strong research background in the field of mRNA storage, translational control, nerve regeneration, drug screening, and peptide based therapy with extensive scientific manuscript and patent writing.
