My research trajectory is mostly related with molecular genetics of bacteria, as Mycobacterium tuberculosis and other non-pathogenic mycobacteria. Our group worked with cell cycle genes transcriptional regulation and cell cycle proteins localization. Although the results on cell cycle proteins localization were not published, we were the first to use antibodies and GFP-protein fusions to locate proteins, as SMC and ParB, in M. tuberculosis and M. smegmatis (results were shown at international meetings and conferences). While working at the Lab. of Molecular Biology, at IVIC (as PhD student, Postdoc and Researcher), I had to standardize most of the techniques used for molecular genetics and fluorescent microscopy. I have always like to do bench work and to implement new protocols in the lab. Also, I was trained to study the virulence of strains with IS6110 insertions near the oriC of M. tuberculosis, in the experimental model of progressive pulmonary tuberculosis in mice, in México. Working in this project involved the manipulation of infected mice, taking samples of blood, lungs and spleen for RNA isolation, bacterial counts or histopathology. Experience with bacterial pathogens allowed me to work as Postdoctoral Fellow at the Microbiology Laboratory at the Glycomics Institute, Griffith University, Australia. Professor Ifor Beacham wanted to create bioluminescent Burkholderia pseudomallei, but was having trouble with stability. Coming from a mycobacterial background, I was familiar with the need to insert genes in the chromosome in order to get stable transformants. Therefore, I suggested the use of chromosomal insertions, instead of a plasmid, to carry the lux cassette. He found out about mini-Tn7 elements and contacted Dr. Herbert Schweizer. Collaboration was established and the use of mini-Tn7 elements to insert the lux cassette into the chromosome of B. pseudomallei made it possible to get a very stable bioluminescent strain that allowed studying the nasal infection route in vivo. The cloning of the lux cassette in the mini-Tn7 was achieved doing a partial digestion of the plasmid carrying it. Expertise in restriction and cloning reactions has allowed me to: isolate promoter regions, express functional proteins fused to tags or GFP, sequence unknown regions of interest, to introduce point mutations and to work with two hybrids systems. More recently, as a Prometeo Researcher in Ecuador, I was asked to improve the microbiological and molecular techniques for diagnosis of bacterial zoonosis, as Salmonellosis and Leptospirosis. They asked if it was possible to identify the serotypes of Salmonella without using antisera? My approach was to use mPCR systems to evaluate the strains isolated during the National Survey of Poultry and Porcine Farms. The systems chosen worked very well for the Salmonella strains isolated from poultry (published results), but did not give much information regarding strains isolated from pigs. Regarding Leptospirosis, several DNA extraction protocols (commercial kits and “in house”) were compared to isolate genomic DNA for PCR detection of Leptospira from bovine urine (manuscript in preparation). Some work related with molecular diagnosis (PCR) of bacterial pathogens of Apis mellifera (bee) was also done. Worked as a full time Professor of Microbiology and Food Microbiology at the Medicine Faculty of the Universidad Central de Venezuela for 9 years. Therefore, I have experience working in molecular biology and fluorescent microscopy, not always with unlimited resources or state of the art technologies, so I have learn to use creativity to solve problems and to approach scientific questions from different angles. Also, I am used to work in multidisciplinary and multicultural groups.
