Geradas artificialmente, em cont\u00ednuo crescimento e instrumentais no diagn\u00f3stico e no tratamento das descobertas, as linhas celulares n\u00e3o precisam de mais introdu\u00e7\u00e3o. Sua categoriza\u00e7\u00e3o prim\u00e1ria considera as caracter\u00edsticas morfol\u00f3gicas, bem como o tecido radicular para isolamento como a base da distin\u00e7\u00e3o. Entretanto, pesquisas em larga escala indicaram diferen\u00e7as acentuadas entre as c\u00e9lulas prim\u00e1rias e as linhas celulares desenvolvidas com base nelas. O que faz destes ingredientes biol\u00f3gicos in vitro tais recursos constitutivos para P&amp;D? Quais s\u00e3o as poss\u00edveis raz\u00f5es para a lacuna em sua aplicabilidade? Estas t\u00eam sido perguntas recorrentes para a ind\u00fastria de Desenvolvimento de Linhas Celulares (CLD). Aqui, falamos sobre as melhorias que foram feitas para racionalizar sua aplicabilidade e reduzir os gargalos. Aqui est\u00e3o algumas aplica\u00e7\u00f5es das linhas de c\u00e9lulas<\/p>\n<h3><span class=\"ez-toc-section\" id=\"Conflict_Surrounding_Application_of_Cell_Lines\"><\/span>Aplica\u00e7\u00e3o de Linhas C\u00e9lulas em Conflito<span class=\"ez-toc-section-end\"><\/span><\/h3>\n<p>Ap\u00f3s d\u00e9cadas de pesquisa em linhas de c\u00e9lulas cont\u00ednuas, que eram consideradas como tendo homogeneidade genot\u00edpica e fenot\u00edpica, os cientistas hoje t\u00eam uma variedade de repert\u00f3rios de linhas celulares para escolher. H\u00e1 3600 linhas de c\u00e9lulas dispon\u00edveis que foram desenvolvidas a partir de 150 esp\u00e9cies biol\u00f3gicas. Come\u00e7ando pela linha de c\u00e9lulas de Sertoli (MSC-1), Hepa1-6, HeLa, e linhas de c\u00e9lulas clonais, a CLD embarcou em uma nova jornada. Os pesquisadores agora s\u00e3o apresentados com diferentes linhas de c\u00e9lulas geradas artificialmente ao n\u00edvel que as c\u00e9lulas-tronco embrion\u00e1rias e do cord\u00e3o umbilical s\u00e3o usadas para a nova CLD.<\/p>\n<p>Por outro lado, os pesquisadores enfrentam os desafios mais dif\u00edceis de lidar com linhas de c\u00e9lulas, pois isso leva a uma anormalidade nos resultados, bem como a um grande desperd\u00edcio de recursos. <a href=\"http:\/\/www.tandfonline.com\/doi\/abs\/10.4161\/spmg.19885\" target=\"_blank\" rel=\"noopener noreferrer\">Gurvinder Kaur et al (2012)<\/a> mostrou que as c\u00e9lulas de Sertoli do rato tinham caracter\u00edsticas imunol\u00f3gicas mais importantes do que as linhas MSC-1. <a href=\"https:\/\/www.researchgate.net\/profile\/Damian_Marshall\/publication\/5775029_The_costs_of_using_unauthenticated_over-passaged_cell_lines_How_much_more_data_do_we_need\/links\/55daf2bf08aed6a199ab08dd.pdf\" target=\"_blank\" rel=\"noopener noreferrer\">Hughes et al (2007)<\/a> indicou a perda de 18-36% de qualidade fenot\u00edpica devido a erros de identifica\u00e7\u00e3o e contamina\u00e7\u00e3o cruzada. Recentemente, <a href=\"https:\/\/phys.org\/news\/2017-04-tools-optimize-cho-cell-lines.html\" target=\"_blank\" rel=\"noopener noreferrer\">C\u00e9lulas do ov\u00e1rio chin\u00eas Hamster (CHOs)<\/a> tornou-se cada vez mais dif\u00edcil de usar para sintetizar uma classe de Biofarmac\u00eauticos conhecida como \"Biol\u00f3gicos\". Consequentemente, as raz\u00f5es para a falta de aplicabilidade das linhas celulares s\u00e3o citadas como frequ\u00eancias mais altas de muta\u00e7\u00f5es nas linhas celulares, sele\u00e7\u00e3o clonal descontrolada dentro das passagens celulares e limita\u00e7\u00f5es na sua transforma\u00e7\u00e3o para <em>in vitro<\/em> modelos.<\/p>\n<h3><span class=\"ez-toc-section\" id=\"Application_of_Cell_Lines\"><\/span>Aplica\u00e7\u00e3o de Linhas Celulares<span class=\"ez-toc-section-end\"><\/span><\/h3>\n<p>No entanto, as linhas celulares continuam sendo os recursos mais rent\u00e1veis e eticamente vi\u00e1veis para a realiza\u00e7\u00e3o de pesquisas cient\u00edficas. Seu desenvolvimento foi idealizado por sua homogeneidade fenot\u00edpica e genot\u00edpica sobre as popula\u00e7\u00f5es celulares recorrentes. Nas \u00faltimas d\u00e9cadas, a viabilidade e a f\u00e1cil disponibilidade das linhas de c\u00e9lulas foram as raz\u00f5es pelas quais quase todos os desenvolvimentos cient\u00edficos foram baseados em linhas de c\u00e9lulas. N\u00e3o \u00e9 de se admirar que elas sejam utilizadas para a produ\u00e7\u00e3o de anticorpos, desenvolvimento de vacinas, testes de drogas, estudos de fun\u00e7\u00e3o gen\u00e9tica, estudos de crescimento de c\u00e9lulas cancer\u00edgenas e desenvolvimento artificial da pele, entre outros.<\/p>\n<p>Novas linhas celulares est\u00e3o sendo continuamente desenvolvidas para encontrar biomarcadores para c\u00e2ncer, doen\u00e7as de pele, disfun\u00e7\u00f5es pulmonares e melhor visualiza\u00e7\u00e3o das c\u00e9lulas tumorais. Como as linhas celulares imitam a homogeneidade celular de quase todos os tecidos humanos e s\u00e3o utilizadas para criar modelos in vitro, a aquisi\u00e7\u00e3o e manuten\u00e7\u00e3o de linhas celulares continua sendo imperativa, com novas diretrizes e protocolos chegando a cada ano pelas empresas CLD. Sigma Aldrich, Thermo Fisher Scientific, Lonza e The Horizon s\u00e3o as empresas que t\u00eam desenvolvido e evolu\u00eddo continuamente as linhas de c\u00e9lulas em termos de variantes citogen\u00e9ticas.<\/p>\n<h3><span class=\"ez-toc-section\" id=\"The_Way_Forward\"><\/span>O caminho para o futuro<span class=\"ez-toc-section-end\"><\/span><\/h3>\n<p>Com os resultados conflitantes do uso de linhas celulares sobre linhas de c\u00e9lulas prim\u00e1rias em m\u00e3os, os pesquisadores t\u00eam a dif\u00edcil tarefa de decidir qual recurso celular deve ser usado. A resposta a isto est\u00e1 em <a href=\"https:\/\/www.thermofisher.com\/in\/en\/home\/life-science\/genome-editing\/genome-engineering-services\/cell-line-engineering-services.html\">M\u00e9todos de engenharia de linha celular (CLE)<\/a> e em cultura celular 3D. Estas novas abordagens s\u00e3o baseadas na obten\u00e7\u00e3o de c\u00e9lulas prim\u00e1rias humanas e\/ou c\u00e9lulas animais a partir de tecidos radiculares e seu cultivo com meios seletivos em todas as 3 dimens\u00f5es, permitindo que as c\u00e9lulas formem agregados em forma de esf\u00e9rulas, conhecidos como esfer\u00f3ides. As condi\u00e7\u00f5es de crescimento para culturas de c\u00e9lulas 3D incluem o uso de tapetes polim\u00e9ricos, por exemplo, tapetes de poliestireno, fibras de ECM \u00e0 base de policarbonato, tapetes de col\u00e1geno e substratos artificialmente projetados in vitro, conhecidos como andaimes.<\/p>\n<p>A CLE \u00e9 uma tecnologia avan\u00e7ada que permite que os CLDs projetem linhas de c\u00e9lulas utilizando os produtos CRISPR. <a href=\"https:\/\/www.thermofisher.com\/blog\/cellculture\/how-can-cell-culture-help-you\/\" target=\"_blank\" rel=\"noopener noreferrer\">Thermo Fisher Scientific<\/a> \u00e9 a engenharia de novas linhas celulares utilizando uma grande variedade de linhas celulares j\u00e1 dispon\u00edveis de acordo com os requisitos do projeto de pesquisa solicitado por seus clientes colaboradores.
